A cathedral school had existed since around 1103, but by the 16th century however, it had declined, and in 1509, a new school was founded by John Colet, Dean of St Paul's Cathedral, on a plot of land to the north of the Cathedral. The original school, was destroyed with the Cathedral in the Great Fire of London in 1666 and rebuilt in 1670. Originally, the school provided education for 153 children as the number 153 has long been associated with the miraculous draught of fishes recorded in St John's Gospel.

It was designed by Robert Hooke and was demolished in 1866. In 1666 the College of Physicians’ home in the City of London was completely destroyed by the Great Fire. The physicians then moved into a spectacularly grand and opulently redesigned home in 1678 in Warwick Lane. It was one of three sites in the City of London near St Paul's Cathedral.
